Purification and component analysis of anthocyanin from blueberry peel

Abstract. The present study is intended to evaluate the adsorption and desorption performance of anthocyanins from blueberry peel using crude extract as raw materials. In addition, the purification conditions of AB-8 macroporous resin to anthocyanin were optimized and the components of anthocyanin were analyzed by using high performance liquid chromatography tandem mass spectrometry in this study. The results showed that the color value of anthocyanin was 52.10 after purification, which proved that AB-8 macroporous resin had a good effect on the anthocyanins purification. The equilibrium time of absorption and desorption were 3 hours and 2 hours, respectively. At the conditions of pH 1.0 and 30, the highest adsorption ability for anthocyanin was achieved, and 40% acidic ethanol solution could be obtained better eluting effect. The best condition of dynamic adsorption was feeding rate 1.0 ml/min, eluant 40% acidic ethanol, desorption rate 1.0 ml/min. Besides, 16 chromatographic peaks were separated from the crude extract and 12 of anthocyanin monomer were determined.
