Impact of brilliant yellow on the synthesis and structure change of Gluconacetobacter xylinus cellulose

Abstract. Bacterial cellulose (BC) was produced by Gluconacetobacter xylinus in the presence of brilliant yellow (BY), fluorescent brightener 28 (FB28) or Pontamine fast scarlet 4B (S4B) under static condition. With increasing concentration of brilliant yellow from 20 μM, 50 μM, 100 μM to 250 μM, the crystallinity of BC decreases considerably from ~ 73% to ~ 18%, compared to ~ 82% of the control bacterial cellulose. In addition, crystal size data shows brilliant yellow impacts crystallization preferentially along the 110 plane. FTIR data indicates that compared to control BC, the brilliant yellow-altered BC contains more cellulose Iβ. Our study shows that BY more efficiently disrupts the crystalline structure than FB28 and S4B, which may due to the planar shape of BY molecules. This study indicates BY is a new powerful tool to disrupt the in vivo assembly of BC.
