ENZYME HYDROLYSIS of WASTE POTATO MASH

The potato is a starchy, tuberous crop vegetable which is one of the carbon sources for ethanol fermentation. However, a pretreatment process is needed to convert starch to fermentable carbon sources, glucose. In order to obtain a maximum glucose conversion, relationship among parameters of the liquefaction and saccharification process were investigated by response surface method. A Box-Behnken Surface Response Method was performed to attain optimum conditionals for high conversion yield. The optimum combination of temperature, dose of enzyme (a-amylase), and amount of potato was 95C, 1 ml enzyme (18.8 mg protein/ml), and 4.04 g dry-weight/100 ml DI water, respectively with a 68.86% loss in dry weight. For saccharification, dose of enzyme, temperature, and saccharification time were examined by creating Box-Behnken Surface Response design and 60C-72 h-0.8 ml (300 Unit/ml) of amyloglucosidase combination was determined as optimum conditionals with 34.9 g/L glucose.
